Has been shown to be activated by activin type I receptor-like kinase-2 (ALK-2) which stimulates heteromerization between SMAD9 and SMAD4. ALK-2 binds TGF-beta, activin and BMP.

Cytoplasm Nucleus In the cytoplasm in the absence of ligand. Migration to the nucleus when complexed with SMAD4.
