Aquaporin-1 is a widely expressed water channel, whose physiological function has been most thoroughly characterized in the kidney.It forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.

It is found in a number of tissues including erythrocytes, renal tubules, retinal pigment epithelium, heart, lung, skeletal muscle, kidney and pancreas.

Aquaporin-1 is assotiate multiple tumor angiogenesis and metastasis, for example , the protein level is up-regualaion in breaste cancer, ovarian cancer, prostate cancer, bladder cancer.

B-type Natriuretic Peptide (BNP) Assay     Background   B-type natriuretic peptide (BNP) is a cardiac neurohormone secreted from membrane granules in the cardiac ventricles as a response to ventricular volume expansion and pressure overload. 1 The natriuretic peptide system allows the heart to participate in the regulation of vascular tone and extracellular volume status. The natriuretic peptide system and the renin angiotensin system counteract each other in arterial pressure regulation. Levels of atrial natiuretic peptide (ANP) and BNP are elevated in cardiac disease states associated with increased ventricular stretch.   The main circulating and storage form of BNP is 32 amino acid peptide with a ring structure. Physiological actions of BNP are mediated through a guanylate cyclase-linked receptor, natriuretic peptide receptor A (NPR-A). BNP produces arterial and venous vasodilation. Clearance of BNP is promoted by a NPR-C receptor which removes it from the circulation and BNP is also degraded through enzymatic cleavage by neutral endopeptidase. BNP levels are reflective of left ventricular diastolic filling pressures and thus correlate with pulmonary capillary wedge pressure.   BNP is stable in whole blood and a portable, 15 minute assay has been developed for measuring BNP in whole blood samples (Triage BNP Test, Biosite Diagnostics). BNP levels have been shown to be elevated in patients with symptomatic left ventricular dysfunction and correlate with New York Heart Association (NYHA) classific ation and prognosis.   Distinguishing congestive heart failure from other causes of dyspnea is of great importance in patients presenting for medical attention with signs and/or symptoms that may or may not represent heart failure. Immunoprecipitation involves the interaction between a protein and its specific antibody, the separation of these immune complexes with Protein A, and the subsequent analysis by SDS-PAGE. This technique provides a rapid and simple means to separate a specific protein from whole cell lysates or culture supernatants. Additionally, one can use immunoprecipitation to confirm the identity or study biochemical characteristics, posttranslational modifications, and expression levels of a protein of interest. The procedure can divided into several stages: 1       Sample preparation 2       Preclearing       Antibody incubation/formation of antibody-antigen complexes       Precipitation       Analysis by SDS-PAGE The protocol below offers a general guideline for immunoprecipitation, optimization may be required for each specific antigen and antibody. The abundance of a given protein n a sample is variable and a critical factor for obtaining desired results. In this protocol, a cell concentration of approximately 10(7) cells/ml is recommended as starting point. Fine adjustments to cell concentrations and volumes of cell lysate used in each immunoprecipitation experiment may be necessary.   The choice of lysis buffer is critical and dependent on the nature of the protein to be studied. NP-40, a non-ionic detergent, is the most commonly used detergent in cell lysis buffers. Increasing the salt concentration, decreasing the detergent concentration, or changing the detergent to Triton X-100, Saponin, Digitonin, CHAPS or others are steps that can be taken to optimize conditions for immunoprecipitation.   The preclearing step is incorporated into the procedure to lower the amount of non-specific contaminants in the cell lysate......
